基底样乳腺癌与ERK信号传导通路关系的研究

摘　　要：目的检测三阴性乳腺癌中CK5/6,EGFR,P-ERK的表达,探讨基底细胞样乳腺癌与ERK/MAPK信号传导通路的关系。方法采用免疫组化法检测CK5/6,EGFR,P-ERK在70例三阴性乳腺癌组织中的表达。结果在(70例)三阴性乳腺癌中,CK5/6与EGFR的阳性表达率分别为68.6%(48/70),57.1%(40/70),共筛出基底细胞样乳腺癌53例。在三阴性乳腺癌中P-ERK在基底细胞样乳腺癌与非基底细胞样乳腺癌的阳性表达率分别为79.2%(42/53),47.1%(8/17),其差异有统计学意义(P<0.05)。CK5/6与EGFR、P-ERK在基底细胞样乳腺癌的表达中具有正相关性(r=0.471;0.389,P<0.05)。结论 CK5/6,EGFR是诊断基底细胞样乳腺癌的重要分子标记物;基底细胞样乳腺癌的发生发展与ERK/MAPK信号传导通路被异常激活有关;CK5/6与EGFR、P-ERK在基底细胞样乳腺癌的表达中具有相关性。Objective To detect the expression of CK5/6,EGFR and P-ERK in triple negative breast carcinoma and to probe into the relationship between basal-like breast carcinoma and ERK/MAPK signal transduction pathway.Methods To detect the expression of CK5/6,EGFR and P-ERK in 70 cases of triple negative breast carcinoma by using immunohistochemistry（IHC）.Results In 70 cases of triple negative carcinoma,the positive rates of CK5/6 and EGFR were 68.6%（48/70）and 57.1%（40/70）respectively,and a total of 53 cases of basal-like breast carcinoma were screened out.In triple negative breast carcinoma,the positive rate of P-ERK in basal-like breast carcinoma and non-basal-like breast carcinoma were 79.2%（42/53）and 47.1%（8/17）respectively,their difference was of statistical significance（P 〈 0.05）.There was a positive correlation among expression of CK5/6,EGFR and P-ERK in basal-like breast carcinoma（r =0.471;0.389,P 〈 0.05）.Conclusion CK5/6 and EGFR can be used as important molecular markers for diagnosing basal-like breast carcinoma;occurrence and development of basal-like breast carcinoma relates to aberrant activation of ERK/MAPK signal transduction pathway;and there is a correlation among expression of CK5/6,EGFR and P-ERK in basal-like breast carcinoma.
