胃癌标本和正常胃组织中P53基因及其下游基因METTL9表达研究

摘　　要：目的联合检测P53及其下游基因METTL9(P53 Activated Protein 1)在胃癌中的表达并探讨其作用及二者之间的关系。方法提取正常胃组织和胃癌及癌旁组织中的RNA,采用荧光定量PCR方法对标本中P53基因及其下游基因METTL9的表达及相对定量结果。结果胃癌组织P53蛋白表达明显低于正常组织和癌旁组织,胃癌旁组织P53蛋白表达明显低于正常组织,胃癌组织和癌旁组织METTL9表达明显高于正常组织,以上结果具有显著性差异(P<0.05),METTL9与P53呈负相关(P<0.05)。结论 P53与METTL9在胃癌组织中高表达且与胃癌的分化程度及转移密切相关,METTL9作为P53的下游基因与P53的表达呈负相关性。Objective United detection P53 and downstream gene in the expression METTL9（P53 Activated Protein 1） gastric P53 and discussed in the stomach METTL9 the role and the relation between the two.Methods Get normal gastric cancer and stomach tissue and the RNA tissue adjacent to cancer,With fluorescent quantitative polymerase chain reaction（PCR） to the specimens METTL9 P53 genes and their downstream gene expression and relative quantitative results.Results Stomach cancer tissue protein expression P53 significantly lower than normal tissue and cancer tissue adjacent,stomach tissue adjacent P53 protein expression significantly lower than normal tissues,Stomach cancer tissue adjacent to organize and express METTL9 significantly higher than normal tissues,The above results have significant differences（P0.05）,METTL9 and P53 have correlation（P0.05）.Conclusion P53 and METTL9 in stomach tissue high expression and the differentiation degree and gastric cancer and metastatic closely related,METTL9 as P53 downstream genes and P53 expression have correlation.
