摘 要:目的研究乳腺癌患者血清蛋白质的变化,建立灵敏度和特异度高的乳腺癌诊断模型。方法利用SELDI-TOFMS技术对194例乳腺癌患者和60例正常人血清蛋白质分别进行检测,IMAC30芯片结合蛋白质后,用蛋白芯片仪读取数据,分析得到区分乳腺癌患者和正常人的树状分类规则,并构建诊断模型。结果 M11368、M7768、M2943等3个蛋白质组成的模型1可将2组准确分组,灵敏度和特异度分别为94.8%(184/194)和96.7%(58/60);以M11368蛋白质单独组成诊断模型2进行区分,灵敏度和特异度分别为89.69%(174/194)和93.33%(56/60)。结论 SELDI-TOFMS技术可以应用于乳腺癌的诊断。Objective To research the serum protein of breast cancer patients and health adults,and establish the diagnostic model of breast cancer with high sensitivity and specificity in application. Methods Samples of sera with 97 breast cancer patients and 30 healthy adults were detected by surface enhanced laser desorption/ionization time of flight mass spectrometry(SELDI-TOF MS).After the surface of chips IMAC30 bind the serum protein,proteinchip reader of ciphergen inc,a part of SELDI-TOF MS,would detect the signs.Then the classification rule of tree was discovered.The model of identification of breast cancer would be constructed by the classification rule of tree. Results At the key M/Z values of M11368,M7768,M2943,which composed MODEL 1,protein contents of the two classes were obviously diferent by the software analysis.And corresponding sensitivity was 94.8%(184/194) and corresponding specificity was 96.7%(58/60).The MODEL 2,composed by M11368 only,corresponding sensitivity was 89.69%(174/194) and corresponding specificity was 93.33%(56/60). Conclusion THE SELDI-TOF MS could be used in the diagnose of breast cancer.
