摘 要:通过正交实验和单因素实验优化了全叶青兰总黄酮类化合物的提取工艺,得出最佳提取工艺为:乙醇浓度为70%,料液比为1∶15,提取温度为55℃,总提取时间2h,其中超声辅助提取时间为30min,总黄酮的提取量为25mg/g左右。对该工艺条件下提取所得到的乙醇提取物采用不同极性的溶剂进行极性部位的初步分离后的定性实验结果表明,各提取部位均含有黄酮类化合物,其中乙醚分离的弱极性部分不含邻二酚羟基的黄酮,而乙酸乙酯提取的部分则含有二氢黄酮或二氢黄酮醇类,全叶青兰中的黄酮类化合物以极性较大的黄酮苷类为主,游离苷元的含量较低。Extraction process of flavonoids from Dracocephalum integrifolium Bge. were obtained combining with orthogonal design methods and single-factor experiment,namely was 70% alcohol( v/v) ,1∶15 ratio of material to liquid,55℃,the total extraction time of 2h,30min treatment by ultrasonic-wave assistant.Based on the conditions, the extraction yield of flavonoids was about 25mg /g.This confirmed that the optimized condition was suitable and feasible.A preliminary separation process was carried out by extracting concentrated ethanol extract with different solvents which have varying polarity.The qualitative analysis results showed that all fractions contained flavonoids. Non-polar fraction obtained from ether extracts contained no ortho-diphenolic hydroxyl flavone and medium-polar fraction obtained from acetic ether extracts contained flavanones or flavanonols.The results indicated that the main portion of the flavonoids from Dracocephalum integrifolium Bge. was flavonoid glycosides,and free flavonoids content were low.
