摘　　要：目的 :了解国人家族性及散发性 AD患者中 2 1号染色体上 APP基因 16、 17外显子的突变情况。方法 :对 3例家族性 AD、 2 6例散发 AD,行静脉取血 ,酚、氯仿抽提法 ,提取基因组 DNA,PCR扩增 APP基因 16外显子及其 5′端 84个及 3′端 86个 bp的内含子部分 ,17外显子及其 5′端 114个及 3′端 69个 bp的内含子部分 ,经玻璃粉末吸附法纯化靶 DNA片段后 ,对全部 AD患者 ,每例样本分别经含与不含甘油的非变性聚丙烯酰胺凝胶电泳 ,行两次聚合酶链式反应 -单链构型多态性 (PCR- SSCP)分析 ,并对家族性及散发 AD患者行 fmol PCR产物 DNA序列分析。结果 :全部 AD患者 PCR- SSCP分析 ,未见异常的泳动条带。对国人健康对照及家族性 AD患者 APP基因 16、 17外显子及扩增片段的内含子部分的 DNA序列结果 ,与 Yoshikai报告的 DNA序列完全一致 ,未见异常的缺失、插入或置换。结论 :综合本文检测结果及国内相关文献 ,可以初步认为国人 AD患者 APP基因突变发生率并不普遍。目的 :了解国人家族性及散发性 AD患者中 2 1号染色体上 APP基因 1 6、 1 7外显子的突变情况。方法 :对 3例家族性 AD、 2 6例散发 AD,行静脉取血 ,酚、氯仿抽提法 ,提取基因组 DNA,PCR扩增 APP基因 1 6外显子及其 5′端 84个及 3′端 86个 bp的内含子?Objective:to investigate the point mutation on 16,17 exon of APP gene in Chinese sporadic and familial patients of Alzheimer’s disease.Method:we extracted the genomic DNA from the blood samples,which came from 22 cases of sporadic AD and 3 cases of familial AD.By the method of PCR,we got the 16,17 exon and their some introns including 84 bp in 5’ terminal,86 bp in 3’terminal of 16 exon,and 114bp in 5’terminal,59bp in 3’terminal of 17 exon.After purified by the glass milk kit,The DNA samples were studied by PCR-SSCP two times:first,gel electrophoresis without glycerol;secend,gel electophoresis containing 10% glycerol.We also sequenced the DNA samples which come from 3 cases of FAD.Results: The whole samples didn’t show any abnormal in PCR-SSCR.The sequencing failed to find any mutation in 3 cases of FAD.Conclusion: According to our and other reports on APP gene of Chinese AD,we can get the primary conlusion that the mutation is uncommon on 16,17 exon of APP gene in Chinese AD.
