多不饱和脂肪酸代谢限速酶基因多态性与吉林省汉族偏执型精神分裂症的病例-对照研究

摘　　要：目的探讨FADS1(Δ-5去饱和酶基因)、FADS2(Δ-6去饱和酶基因)和ELOVL2(长链脂肪酸延长酶基因)多态性在中国吉林省汉族偏执型精神分裂症发生中的作用。方法研究对象为吉林省100例偏执型精神分裂症患者和95例健康人,采用PCR-RFLP方法检测FADS1、FADS2和ELOVL2基因3个SNPs(单核苷酸多态性)位点的基因型。结果 FADS1基因rs174556位点和FADS2基因rs174617位点基因型在病例组与对照组的分布无显著差异(P>0.05);ELOVL2基因rs3756963位点基因型在病例组与对照组的分布有统计学差异(P<0.05),T等位基因携带者在病例组的分布高于对照组(P<0.05)。trans-phase基因联合作用分析发现包含rs3756963位点T/T的基因型组合在病例组分布多于对照组(P<0.05)。结论 FADS1基因rs174556位点和FADS2基因rs174617位点与偏执型精神分裂症可能不相关,ELOVL2基因rs3756963位点多态性可能与偏执型精神分裂症的发生相关。Objective To investigate an association between gene polymorphisms of FADS1（fatty acid desaturase 1）,FADS2（fatty acid desaturase 2） and ELOVL2（elongation of very long chain fatty acids（FEN1/Elo2,SUR4/Elo3,yeast）-like 2） and paranoid schizophrenia of the Han ethnicity in Jilin province of China.Methods We genotyped 3 single nucleotide polymorphisms from 3 candidate genes in 100 paranoid schizophrenia cases and 95 healthy controls using polymerase chain reaction-restriction fragment length polymorphism method.All samples are from Han ethnicity in Jilin province of China.Results The genotype distributions of rs174556 in FADS1 gene and rs174617 in FADS2 gene showed no significant difference between case and control groups（P 0.05）.The genotype distribution of rs3756963 in ELOVL2 gene showed significant difference between case group and control group（P 0.05）.The distribution proportion of allele T carriers in case group was higher than that in control group.Trans-phase gene interaction analysis showed that the distribution proportion of combined genotypes included rs3756963（T/T） was higher in case group than that in control group（P 0.05）.Conclusion rs174556 in FADS1 gene and rs174617 in FADS2 gene may not be associated with paranoid schizophrenia.rs3756963 in ELOVL2 gene may be associated with paranoid schizophrenia.
