摘  要:目的 观察丹参多酚酸盐对局灶性脑缺血再灌注损伤后大鼠脑组织内CAT、XO和NO含量的影响,探讨其脑保护作用机制.方法 线栓法制作大鼠中动脉缺血再灌注损伤模型.SD大鼠随机分为正常对照组,缺血再灌注组,丹参多酚酸盐低剂量治疗组(10mg/kg)和丹参多酚酸盐高剂量治疗组(30mg/kg).各组动物按要求给药,在预定时间进行神经损伤行为学评分、脑梗死体积及脑组织内过氧化氢酶(CAT)、黄嘌呤氧化酶(XO)活性和一氧化氮(NO)含量的测定.结果 与缺廊再灌注组相比,丹参多酚酸盐治疗组大鼠神经损伤行为学评分及脑梗死体积减少.脑组织内CAT活性升高,XO活性降低,NO含最下降,均具有显著性意义,且呈剂量依赖性.结论 丹参多酚酸盐可通过抑制自由基引起的脂质过氧化反应,保护抗氧化酶的活性而介导神经保护作用.Objective To detect the protective effect of salvianolate on the focal cerebral ischemia- reperfusion injury in rats and to reveal the mechanism. Methods 48 SD rats were divided into four groups randomly, normal control group,ischemia-reperfusion group,salvianolate(10mg/kg) group and salvianolate (30mg/ kg) group,each group contain 12 rats.We used the Longa method to establish the MCAO model.The salvianolate at different dose were injected intraperitoneally at different time point according to the test design.The brain infarct volume by TTC stain and behavior scores by Bederson' s methods were also determined in order to evaluate the protective effect.Also we use assay kit to determine the activities of CAT and XO,content of NO in each group. Results Salvianolate can reduce both the brain infarct volume and behavior score.The effect of salvianolate(30mg/ kg) is stronger than that of salvianolate(l 0mg/kg).The activities of CAT of brain tissue in the ischemia control group,salvianolate(10mg/kg) group and salvianolate(30mg/kg) group are much lower than those in normal control group.But the salvianolate can inhibit this decrease.The activities of XO and the content of NO of brain tissue in the ischemia control group,salvianolate(10mg/kg) group and salvianolate(30mg/kg) group are much higher than those in normal control group.The salvianolate can inhibit this increase. Conclusion The salvianolate plays neuroprotective effect maybe through modulating the imbalance of oxidant and antioxidant inducing by the cerebral ischemia-reperfusion.
