摘 要:目的:探讨表没食子儿茶素没食子酸酯(EGCG)对鼻咽癌细胞增殖、凋亡及生存素(survivin)和血管内皮生长因子(VEGF)的影响。方法:体外培养鼻咽癌细胞,以不同质量浓度EGCG处理,采用MTT法检测细胞增殖抑制作用,Hoechst33258染色观察凋亡细胞并计算细胞凋亡率,流式细胞仪检测细胞周期变化,RT-PCR检测survivin和VEGF的表达变化。结果:EGCG处理后,CNE-2细胞增殖明显受到抑制,呈时间和剂量依赖性;CNE-2细胞凋亡率显著升高;CNE-2细胞周期阻滞于G1/G0,表现为浓度依赖性;survivin和VEGF的表达明显下调,随EGCG浓度的增加有下降趋势。结论:EGCG能抑制鼻咽癌细胞增殖、促进其凋亡;机制可能与EGCG下调survivn和VEGF的表达有关。Objective To study the effects of epigallocatechin-3-gallate (EGCG) on the proliferation, apoptosis and expressions of survivin and VEGF in nasopharyngeal carcinoma cell line CNE-2. Methods Nasopharyngeal carcinoma cells was cultured in vitro, after treated with various concentrations of EGCG, the cell proliferation ability was detected by MTF; Apoptosis of cells was observed by Hoechst staining with apoptosis rate counted;Flow cytometry was used to analyze the changes of cell cycle. RT-PCR was applied to detect the expression of survivio and VEGF. Results EGCG could effectively inhibit the growth of CNE-2 cells, arrest the cells in GI phase in a dose-dependent manner and induce cell apoptosis. Furthermore, EGCG could down-regulate the expressions of survivin and VEGF after treated with EGCG. Conclusions EGCG could significiantly suppress the proliferation and induce apoptosis in uasopharyngeal carcinoma cell line CNE-2; The possible mechanisms may be involved with decreasing the expressions of survivin and VEGF.
