摘　　要：以光敏生物素分别标记含目的基因的L、E选择素重组质粒,用DNaseI随机消化为100bp大小的片段作为探针,用于检测细胞中L选择素或E选择素mRNA的表达水平。结果显示,经TNFα和IL1刺激后的脐带内皮细胞中人E选择素mRNA有较强的表达信号;Raji,HL60,KGI和U937细胞,均有L选择素的表达。这些结果为L、E选择素探针用于临床检测和诊断奠定了基础。Adhesionmoleculesisakindofmoleculeswhichmediatetheinterreactionofcellwithothercellandtheextracellular matrix .L ,E selectin ,asamemberofadhesionmoleculefamily ,hasanimportantfunctionsininflammatoryresponse,lym phocytehomingandtumormetastasis.TherecombinantplasmidswithL orE selectingenearelabelledwithphoto biotinand thendigestedwithDNaseI.TheinsituhybridizationtechniquewasusedtoinvestigatetheL orE selectinmRNAlevelsindif ferentsamples.TheresultsshowedthattherewasstrongersignalofE selectinmRNAexpressioninumbilicalcordendo thelial cellsstimulatedwithTNF αandIL 1,L selectinmRNAexpressioninRaji,HL60 ,KG IandU93 7cells.Theseresultslay thefoundationforclinicaldetectionanddiagnosisofL andE selectins.
