摘 要:目的检测胰腺癌组织中糖酵解关键酶活性及糖酵解产物含量,分析糖酵解与胰腺癌组织生物学行为的关系。方法收集21例胰腺癌组织标本和12例胰岛素瘤等良性病变胰腺组织,制备组织匀浆,用化学比色法检测糖酵解关键酶己糖激酶(hexokinase,HK)、丙酮酸激酶(pyruvate kinase,PK)活性及糖酵解产物乳酸含量。结果与正常胰腺组织相比,癌组织中HK、PK活性和乳酸含量明显升高,差异有统计学意义(P<0.05)。结论胰腺癌有氧氧化功能降低,糖酵解水平增强,特异性抑制糖酵解可能成为胰腺癌新的治疗策略。Objective To test the activity of hexokinase(HK),pyruvate kinase(PK) and the lactate content in pancreatic carcinoma tissue,and to explore the relation between biological behavior of pancreatic cancer and glycolysis.Methods We collected the tissue samples from 21 cases of pancreatic adenocarcinoma and 11 cases of benign lesions such as insulinoma.The total activity of HK,PK and the content of lactate were tested by chemical colorimetry respectively.Results The activity of HK,PK and the lactate content were significantly higher than that of the non-cancerous tissue(P〈0.05).Conclusion Inhibition the glycolysis may be a new therapeutic method on pancreatic cancer treatment.
