摘 要:目的探讨微卫星杂合性缺失(LOH)在复发性喉鳞状细胞癌患者原发癌的特征及其临床意义。方法选取染色体3p、5p和9p上3个多态性微卫星位点D3S1234、D5S592、D9S171,利用PCR-SSCP银染技术,对已复发的31例原发喉鳞状细胞癌组织及相应癌旁组织进行LOH分析。结果 3个微卫星标记物LOH发生率分别为48.3%(15/31),45.2%(14/31),61.3%(19/31)。LOH的检出率与第一次术后至复发时间(复发时间)呈负相关,即检出患者术后复发时间短,未检出患者术后复发时间长,两组间差异有统计学意义(P<0.05)。结论微卫星杂合性缺失与术后复发时间呈负相关,可作为喉癌预测术后复发的参考标志,有效指导术后临床治疗方案的选择。Objective To study the characteristic and significance of microsatellite loss of heterozygosity(LOH)in primary foci of recurrence laryngeal squamous cell carcinoma.Methods Allelic deletions analysis was performed on primary carcinoma and adjacent tissues in 31 cases of recurrence laryngeal squamous cell carcinoma with 3 polymorphic markers(D3S1234,D5S592 and D9S171)on 3p,5p and 9p chromosomal arms by polymerase chain reaction-single strand conformational polymorphism(PCR-SSCP)-silver staining.Results Occurrence rates of LOH in the three markers were 48.3%(15/31),45.2%(14/31)and 61.3%(19/31),respectively.The patients with LOH detected had shorter recurrence time than those with no LOH detected.A statistical significance was found between the two groups(P〈0.05).Conclusions There is a negative correlation between LOH and recurrence time.LOH has a predictive value with respect to post-operative relapse in laryngeal cancer patients.It is effectively instructive to choose the best clinical treatment protocol after operation.
