摘 要:对影响玉米胚芽超氧化物歧化酶(SOD)提取的浸泡条件进行研究。利用单因素法优化的最佳浸泡工艺条件为:100g玉米中加入200mL0.05mol/L磷酸缓冲液,40℃浸泡玉米36h,淋干后分离胚芽与胚乳,再加入胚芽质量2倍的0.05mol/L磷酸缓冲溶液浸提1h,离心得SOD提取液,其总酶活为22549.0U;而未浸泡的玉米胚芽SOD总酶活为2125.3U。由此可知,浸泡对玉米胚芽SOD提取的影响非常显著。The effects of the steeping on the extraction of superoxide dismutase(SOD) from embryo were studied.The Steeping conditions of maize were optimized by single-factor design before extraction of SOD from embryo.The optimal extracting conditions of SOD from embryo were that 100 g maize was dipped with 200 mL 0.05 mol/L phosphate buffer at 40 ℃ for 36 h and then filtered.The 0.05 mol/L phosphate buffer was added as much as twice embryo to smash it after embryo and endosperm separated,stirring for 1 h,and extracts of SOD were obtained by centrifugation,whose total SOD activities were 22 549.0 U;however,the total SOD activities of embryo without steeping were 2 125.3U.There was a large influence of steeping on extraction of SOD from corn.
