摘  要:目的:探讨特异性siRNA对卵巢癌SKOV3细胞中肿瘤-睾丸抗原OY-TES-1蛋白表达的影响。方法:针对OY-TES-1基因设计、化学方法合成的siRNA通过脂质体转染体外培养的卵巢癌SKOV3细胞,在转染后24、36、48、72 h,通过免疫细胞化学检测OY-TES-1蛋白的表达水平。结果:靶向OY-TES-1基因siRNA转染SKOV3细胞后,OY-TES-1蛋白的表达明显降低,其抑制作用在转染后24 h出现,转染后36 h几乎完全被抑制,转染后72 h开始出现RNAi衰减现象。结论:OY-TES-1特异性siRNA(1 486~1 504)序列可在体外有效地干扰SKOV3细胞中OY-TES-1基因的表达。Objective:To explore the effect of specific siRNA on OY-TES-1 protein expression in ovarian cancer SKOV3 cells. Methods:siRNA targeting to OY-TES-1 gene was synthetized chemically,then ovarian cancer SKOV3 cells were cultured in vitro after liposome transfection by siRNA,at 24,36,48 and 72 hours after transfection,immunocytochemistry was used to detect the expression level of OY-TES-1 protein. Results:After transfecting ovarian cancer SKOV3 cells by siRNA targeting to OY-TES-1 gene,the expression of OY-TES-1 protein decreased,the inhibiting effect appeared at 24 hours after transfection.At 36 hours after transfection,the expression of OY-TES-1 protein was almost inhibited,at 72 hours after transfection,the attenuation of RNA interfering appeared. Conclusion:The specific siRNA sequence(1 486~1 504) of OY-TES-1 protein can interfere the expression of OY-TES-1 gene in SKOV3 cells effectively
