eNOS G894T基因多态性与胃癌发生发展关系的研究

摘　　要：目的:探讨内皮型一氧化氮合酶(Endothelial NOS,eNOS)基因多态性与胃癌发生发展的关系。方法:采用PCR-RFLP技术,检测福建地区汉族324例胃癌患者(病例组)和200例健康对照者(对照组)eNOS基因G894T多态性。结果:(1)在病例组和对照组中e,NOS G894T SNP基因型频率G/G分别为76.5%(248/324)和73.0%(146/200),G/T+T/T分别为23.5%(76/324)和27.0%(54/200),等位基因频率G分别为88.0%(570/648)和86.3%(345/400),T分别为12.0%(78/648)和13.7%(55/400)。eNOS G894T SNP基因型和等位基因频率在病例组与对照组的分布差异无统计学意义(均P>0.05);弥漫型胃癌和肠型胃癌分别与对照组比较,任一基因型或等位基因亦未增加患癌风险(均P>0.05)。(2)eNOS G894T基因型频率分布与胃癌的临床病理特征无明显关系(均P>0.05)。结论:福建地区汉族人群eNOS基因G894T多态性与胃癌的发生和发展无关。Objective：To investigate the association of eNOS gene G894T polymorphism and the development and progression of gastric cancer.Methods：PCR-RFLP method was used to determine the genotypes of eNOS G894T gene polymorphism in 324 gastric cancer patients and 200 cancer-free controls in Han Population of Fujian Province.Results：（1）In the case and control group,G/G genotype frequencies were 76.5%（248/324） and 73.0%（146/200）respectively;G/T＋T/T genotype frequencies were 23.5%（76/324） and 27.0%（54/200）respectively;G allele frequencies were 88.0%（570/648）and 86.3%（345/400） respectively;T allele frequencies were 12.0%（78/648）and 13.7%（55/400）respectively.The comparisons between the case group and control group showed no significant difference in genotype and allele frequencies（all P0.05）;When case group was subdivided as diffuse-type and intestinal-type,the comparisons between the two subgroups and control group respectively also showed that not any genotype or allele increased the risk of gastric cancer（all P0.05）.（2）No statistically significant correlations existed between eNOS G894T polymorphism and several clinicopathological features of gastric cancer（all P0.05）.Conclusion：No association is found between eNOS gene G894T polymorphism and the development and progression of gastric cancer in Han Population of Fujian Province.
