摘  要:目的:对HPV-16阳性的H8细胞株(宫颈上皮永生化细胞)和Caski细胞株(宫颈癌细胞)胞浆蛋白进行双向电泳和质谱分析,寻找与宫颈癌相关的特异性差异蛋白质。方法:采用双向凝胶电泳(2-dimensronal gel electrophoresis,2-DE)和基质辅助激光解吸飞行时间串联质谱(Matrix-assisted laser desorption/ionization time-of-flight tandem mass spectrometry,MALDI-TOF-MS/MS)以及SWISS-PROT数据库,对H8和Caski细胞胞浆蛋白进行比较,找出差异蛋白质。结果:通过对宫颈永生化细胞和癌细胞胞浆蛋白2-DE图谱比较和质谱分析,初步鉴定出6个差异蛋白,它们分别为STK3、TRAF5、S100PI、KKb、CDK6、TAF2,其中S100P、IKKb、CDK6、TAF2、TRAF5这5种蛋白质在宫颈癌细胞胞浆中高表达,STK3蛋白质在宫颈癌细胞胞浆中表达缺失。结论:初步发现STK3、TRAF5、S100PI、KKb、CDK6、TAF2蛋白质在宫颈癌前病变和宫颈癌细胞胞浆中存在差异表达,通过对其功能的了解研究为宫颈癌前病变向癌转变机制的探讨、特异性肿瘤标志物的寻找提供实验依据。Objective:To identify differentially expressed protein spots of cytoplasm from H8 cells(human immortalized endocervical cell) and Caski(cervical cancer cell) associated with cervical carcinogenesis by two-dimensional electrophoresis(2-DE) analysis.Methods:The cytoplasmic proteins of H8 cell and Caski cell were separated by means of two-dimensional gel electrophoresis,then the selected differential protein spots were analyzed by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF-MS) and SWISS-PROT database.Results:6 differentially expressed protein spots were successfully identified,among which,S100P,IKKb(Inhibitor of nuclear factor kappa-B kinase subunit beta),CDK6(Cell division protein kinase 6),TAF2(Transcription initiation factor TFIID subunit 2),TRAF5(TNF receptor-associated factor 5) were significantly up-regulated in cytoplasmic proteins in Caski cell.STK3(Serine/threonine-protein kinase 3) deletion was found in cytoplasmic proteins in Caski cell.Conclusion:The differential expressions of proteins of cytoplasm may be valuable for studying mechanisms of cervical carcinogenesis,or as diagnostic markers and therapeutic targets for cervical cancer.
