摘 要:目的探讨Fasudil对实验性自身免疫性脑脊髓炎(EAE)小鼠脊髓内细胞及血管内皮细胞ROCK-Ⅱ表达的影响。方法雌性C57BL/6小鼠随机分为佐剂组、EAE组和Fasudil干预组。EAE和Fasudil组小鼠用髓鞘少突胶质细胞糖蛋白35-55(MOG35-55)进行免疫,佐剂组小鼠以生理盐水代替MOG35-55作为EAE的对照。Fasudil干预组于免疫后第7d腹腔给予Fasudil(50mg/kg/d),连续14d。佐剂组和EAE组给与等量生理盐水作为对照。免疫后隔天观察各组小鼠临床评分和体重变化。在免疫4周后处死动物,取小鼠脊髓腰膨大处,进行HE和免疫荧光染色,观察各组小鼠脊髓的病理变化与ROCK-Ⅱ的表达情况。结果 Fasudil能够改善EAE的临床症状(P<0.05),减轻炎性细胞浸润(P<0.05),同时减少EAE小鼠脊髓及其血管内皮细胞ROCK-Ⅱ的表达。结论 EAE小鼠脊髓及其血管内皮细胞ROCK-Ⅱ的表达明显上调,推测可能与炎性细胞浸润和疾病的发生发展有关。Fasudil可以改善EAE的临床症状,可能与脊髓及其血管内皮细胞ROCK-Ⅱ降低,及由此引起的炎性细胞浸润减少有关。Objective To investigate the effect of Fasudil on expression of ROCK-Ⅱ in spinal cords and vascular endothelial cells in experimental autoimmune encephalomyelitis mice.Methods Female C57BL /6 mice were randomly divided into adjuvant group,EAE group and Fasudil-treated group.EAE group was set up as Fasudil-treated group control,while adjuvant group was performed as EAE group control.Fasudil was intraperitoneally injected in the dosage of 50mg·kg-1·d-1 on day 7 post-immunization(p.i.) for 14 consecutive days.Mice in EAE and adjuvant group were injected with saline.Clinical signs were evaluated and body weight was recorded every other day.Mice were sacrificed on days 28-30p.i.,and the pathological changes in spinal cords were detected by HE staining.The expression of ROCK-Ⅱ in spinal cords was detected by immunofluorescence staining.Results The clinical symptom and the inflammatory infiltration of spinal cords in Fasudil-treated mice were significantly ameliorated compared with those of EAE mice(P0.05).After the intervention of Fasudil,the expression of ROCK-Ⅱin spinal cords was decreased.Conclusion The expression of ROCK-Ⅱ was significantly upregulated in EAE mice,while treatment of Fasudil improved the clinical symptoms of EAE mice,which perhaps relates to the inhibition of ROCK-Ⅱand the decrease of inflammatory infiltration.
