摘  要:提取了一株芽孢杆菌BLYS-1蛋白酶,探讨了pH值、温度、有机溶剂、金属离子、氧化剂和EDTA等对其的影响.研究发现,该芽孢杆菌蛋白酶具有一定酸碱耐受性和抗氧化性,在pH值58时活性较高,最适宜pH值为7,最佳反应温度为50℃,70%饱和度的硫酸铵可使其完全沉淀.吐温-20和异丙醇、变性剂SDS、金属离子Sn^2+、Ca^2+、Ni^+对蛋白酶抑制作用较大,Mg^2+、Zn^2+等对其有促进作用.以干酪素为底物,测得该酶动力学参数Km为2.53 g/L,Vmax为6.67μg/s.We extracted a protease from a Bacillus strain BLYS-1,and discussed the effects of different factors,such as pH value,temperature,organic solvents,metal ions,oxidant and EDTA,on the protease.The results showed that the protease from Bacillus strain BLYS-1 had resistance to acid,alkali and oxidant,and had higher activity in the pH range of 5 to 8;the best pH value was 7; the best reaction temperature was 50 ℃;and the protease was precipitated completely by ammonium sulfate with the 70% degree of saturation.The organic solvents,such as tween-20 and isopropyl alcohol,the denaturant SDS,and the metal ions,such as Sn^2+,Ca^2+,Ni^+,had higher effect in inhibiting the protease,but Mg^2+ and Zn^2+ had the promotion effect.Based on the casein substrate,the paper determined the kinetic parameter Km of the protease was 2.53 g/L,and Vmax 6.67 μg/s.
