摘  要:目的观察大蒜素和多西他赛(DXT)单用及联合应用对人骨肉瘤细胞株MG-63增殖抑制作用。方法采用MTT法检测细胞毒性作用。倒置相差显微镜观察细胞形态改变,流式细胞仪检测药物对细胞周期的影响,并检测凋亡率。结果大蒜素和DXT单用及联合用药,在一定浓度范围内均对骨肉瘤细胞MG-63有增殖抑制作用和诱导凋亡作用,其作用与剂量呈正相关。联合用药组作用明显强于单独用药组(P<0.05)。大蒜素和DXT单独及联合作用于人骨肉瘤细胞株MG-63,与细胞生长分裂关系较密切的G2/M期占细胞周期的比例有明显升高,且联合用药的升高作用更明显(P<0.01)。结论大蒜素和DXT均可将骨肉瘤细胞株阻滞于G2/M期,两药联合可能具有协同抗肿瘤作用。Objective Observation of the inhibition function of Allicin and Docetaxel(DXT) as monotherapy and in combination on human osteosarcoma cell line MG-63 proliferation.Methods Using MTT method to detect cytotoxicity.To observe the cellular morphological changes by Inverted phase contrast microscope.To analyze the impact of drugs on cell cycle by flow cytometry and detection of apoptosis.Results Allicin and Docetaxel either alone or combined inhibited the proliferation and induced apoptosis of osteosarcoma cells MG-63.function positively correlated with the dose,and the combination of two drugs was obviously more effective than that of the drugs alone(P0.05) Observation of Allicin and DXT as monotherapy and in combination on human osteosarcoma cell line MG-63 cycle: percentages of phase G2/M cells which are closely related with cell growth and division significantly increased,which is even more significant in combination therapy(P0.01).Conclusions Allicin and docetaxel,used to ostersarcoma cells MG-63,can block them at G2/M phase and the two drugs may have combined synergistic anti-tumor effect.
