摘  要:目的探讨RhoC-shRNA对结肠癌细胞生物学行为的影响。方法构建RhoC-shRNA表达载体,以脂质体转染法将之稳定转染入HT-29结肠癌细胞株,Western blot检测转染细胞中RhoC蛋白表达的抑制情况,观察转染前后结肠癌细胞周期、凋亡和侵袭能力的变化。结果成功构建RhoC-shRNA真核表达载体,并且转染入结肠癌细胞后,细胞中RhoC蛋白表达水平明显受到抑制,并且抑制细胞增殖、促进细胞凋亡,降低肿瘤细胞侵袭能力。结论下调RhoC蛋白的表达可抑制结肠癌细胞HT-29的体外增殖和侵袭能力。Objective To investigate the effects of shRNA inhibition of RhoC expression on the biological behavior of HT-29 cells.Methods Recombinant expression plasmid pSihRhoC targeting human RhoC gene was constructed.HT-29 cells were transfected with pSihRhoC.The cell cycle,invasiveness and apoptosis of human colon carcinoma cell line HT-29 were determined by flow cytometry,matrigel invasion assays and Hoechst 33258.Results Recombinant plasmid pSihRhoC was constructed successfully.The expression levels of RhoC protein were down-regulated in HT-29 transfected with pSihRhoC,and inhibited cell proliferation,promoted apoptosis and reduced tumor cell invasion.Conclusion The Recombinant plasmid RhoC-shRNA inhibited the HT-29 colon cancer cell proliferation and invasion in vitro.Therefore,our study provided a novel way for treatment of colon cancer.
