摘  要:应用Plackett-Burman、Box-Behnken设计实验结合期望函数优化蛹虫草固体栽培培养基。以蛹虫草子实体生长周期、子实体产量、多糖、蛋白、腺苷和甘露醇的含量作为考察指标,采用期望函数将多个考察指标综合为一个考察指标——期望值D,对蛹虫草诱变菌株N165固体培养的营养液成分进行优化。蛹虫草诱变菌株N165固体培养基的营养液的最佳配方为:酵母浸粉10.33g/L,蔗糖27.24g/L,KH2PO45.60g/L,蛋白胨5.00g/L,MgSO.47H2O1.00g/L,ZnCl20.011g/L,维生素B1 0.05g/L和(NH4)2SO421.00g/L。The solid culture medium of Cordyceps militaris was optimized with Plackett-Burman and Box-Behnken designs combined with desirability function. Desirability function was used to construct a comprehensive value D, which was an integration index of the growth cycle of fruiting body, the yield of fruiting body, and the contents of polysaccharide, protein, adenosine and mannitol, for the optimization of solid culture medium of C. militaris mutant N165. The optimal medium formula for C. militaris N165 consisted of 10.33 g/L yeast extract,27.24g/L sucrose,5.60g/L KH2 PO4 ,5.00g/L peptone, 1.00g/L MgSO4.7H2 0,0.011 g/L ZnC12 ,0.05g/L VB1 and 21.00g/L (NH4 )2 SO4.
