细胞周期素D1蛋白和mRNA在胆管癌中的表达及其意义

摘　　要：目的检测胆管癌和癌旁0．5cm胆管组织及手术切缘的正常胆管组织细胞周期素D1（CyclinD1）蛋白及mRNA的表达，探讨其在胆管癌发生发展中的作用。方法应用流式细胞分析术检测42例胆管癌和20例正常胆管黏膜组织CyclinD1蛋白的表达，同时应用逆转录-聚合酶链反应（RT—PCR）检测42例术中所取的新鲜的胆管癌、17例同个体癌旁胆管黏膜和20例正常胆管组织中CyclinD1mRNA表达，并与临床资料进行相关分析。结果正常胆管组织、癌旁组织、胆管癌组织中CyclinD1mRNA相对表达量分别为0．5845±0．0310、0．5608±0．0577、0．6288±0．0942．CyclinD1mRNA在3组之间表达呈上升趋势（P〈0．05）。CyclinD1蛋白表达趋势同其相对应的基因表达趋势一致，即CyclinD1的mRNA在胆管癌组织中高表达（P〈0．05）。结论CyclinD1基因转录和蛋白可能参与了胆管癌的发生发展过程。Objective To detect the expression of Cyclin D1 protein and gene in cholangioncarcinoma and to elucidate its role. Methods The expression of Cyelin D1 protein was detected by flow cytom- etry （FCM） in 42 cases of cholangionearcinoma and 20 cases of normal bile duct, and the expression of Cyelin D1 gene was assayed in 42 eases of cholangioncarcinoma, 17 cases of tissues adjacent to cholangioncarcinoma and 20 cases of normal bile duct by reverse transcription-polynaerase chain reaction （RT-PCR）. The correlation was analyzed statistically between the expression of Cyclin D1 protein and mRNA and clini- copathological parameters in cholangioncarcinoma. Results The expression of Cyelin D1 mRNA in normal bile duct, tissues adjacent to cholangioncarcinoma, and cholangioncarcinoma was 0. 5845 ± O. 0310, 0. 5608 ±0. 0577, 0. 6288± 0. 0942 respectively. The expression of Cyclin D1 mRNA was higher in cholangioncarcinoma than in others （ P 〈 0. 05 ）. The protein expression of Cyclin D1 was higher in cholan- gioncarcinoma than in normal bile duct tissue （ P 〈 0. 05 ）. Conclusion Cyelin D1 gene transcription and protein expression is mostly likely involved in the proliferation and malignant transformation of bile epitheli- al cells and then malignant progression of bile duct carcinomas. The detection of Cyclin D1 expression may elucidate the carcinogenesis of bile duct.
