摘 要:目的:探讨小鼠胚胎发育过程早期造血组织中Notch4信号的基因与蛋白的表达情况。方法:采用PT-PCR和Western blot方法,对小鼠胚胎孕9.5 d(E9.5)、E10.5、E11.5、E12.5胚胎的主动脉-性腺-中肾(Aorta-gonad-mesonephros,AGM)区和胎肝中Notch4信号基因和蛋白的表达进行检测。结果:2组小鼠早期造血相关组织的Notch4 mRNA与蛋白表达,在AGM区E10.5组较其它组明显增高(P<0.05);在AGM区和胚胎中都是随着孕龄的增大而逐渐减低(P<0.01)。AGM区与胚胎中比较,在9.5 d和12.5 d有统计学意义(P<0.05);在10.5 d与11.5 d无统计学意义(P>0.05)。PT-PCR结果显示表达规律相同。结论:Notch4信号在小鼠胚胎早期造血组织中有表达,但其功能有待进一步研究。Objective:To investigate the expression of Notch4 gene and protein in the hematopoietic system in mouse early embryos. Methods : The expression of Notch4 mRNA and protein of the Hematopoietic system form the mouse embryonic aorta-gonad-mesonephros ( AGM )region and fetal liver at embryonic day ( E )9.5, El0.5, E11.5, E12.5 were detected by RT-PCR and Western blot techniques. Results:In two groups of mouse early relevant hematopoietic tissue,Notch4 mRNA and protein expression of El0.5 group were significantly higher than other groups ( P〈0.05 ) in the AGM area, and decreased with the pregnancy in both the AGM and fetal hver ( P〈0.0I ). There was statistical difference at embryonic day ( E ) 9.5 and 12.5 in the area comparison between AGM and fetal live ( P〈0.05 ). There was no statistical significance ( P 〉0.05 ) at embryonic day ( E ) 10.5 and 11.5. PT - PCR showed the same expression rules. Conclusion: The Notch4 signals are expressed in early hematopoietic tissue of mouse embryos, but its function is to be further researched.
