摘 要:对青岛近海及威海近海海水和海洋沉积物中的细菌进行了分离和纯化.通过苯酚硫酸法对分离得到的海洋细菌在液体培养条件下的产胞外多糖能力进行了初步的测定,筛选获得了13株生产胞外多糖能力较高的海洋细菌.其中菌株0417胞外多糖产量最高,可达144.212μg/ml.观察细菌的菌落形态特点,并且利用PCR技术对其中的7株进行16SrRNA基因的克隆,将获得序列进行分子鉴定.经序列比对鉴定出这些细菌分属于Bacillus,Aheromonas,Pseudoalteromonas三个属,由此构建系统发育树.The thirteen marine bacteria strains which produced extracellular polysaccharide were isolated from the seawater samples from Qingdao and Weihai offshore. The seven strains were chosen for further work. The 16S rRNA genes were cloned and sequenced to identify the strains. The results show that most of these bacteria are Alteromonas ,Pseudoalteromonas and Bacillus. The yield of polysaccharide in liquid medium for the 13 strains were determined. The highest yield of polysaccharide reached to 144. 212 μg/ml by strain 0417.
