摘 要:目的观察表没食子儿茶素没食子酸酯(EGCG)对胃癌BGC-823细胞增殖和VEGF表达的影响。方法用5、10、20、40、80μmol/L的EGCG培养BGC-823细胞48 h,采用MTT法测定BGC-823细胞增殖抑制率,并计算IC50。用12、24、48μmol/L EGCG处理BGC-823细胞24 h,用Trizol法提取RNA,采用实时定量PCR法检测VEGFmRNA;同时收集细胞上清液,用ELISA法检测VEGF蛋白。结果 BGC-823细胞增殖抑制率与EGCG浓度呈正相关(r=0.977、P<0.05)。EGCG抑制BGC-823细胞增殖的IC50为(47.94±3.26)μmol/L。以12、24、48μmol/LEGCG处理BGC-823细胞24 h,BGC-823细胞中VEGF mRNA相对表达量分别为0.63±0.05、0.25±0.02、0.16±0.01,BGC-823细胞中VEGF mRNA相对表达量与EGCG浓度呈负相关(r=-0.855、P<0.05);上清液中VEGF水平为(677.32±41.08)、(545.16±28.17)、(256.06±11.02)ng/L,上清液中VEGF水平与EGCG浓度呈负相关(r=-0.991、P<0.05)。结论 EGCG可以抑制胃癌细胞的增殖,并且可以下调VEGF mRNA的表达,减少VEGF的分泌。Objective To observe the effects of(-)-epigallocatechin-3-gallate(EGCG) on BGC-823 gastric cancer cell proliferation and VEGF expression.Methods BGC-823 cells were treated for 48 h with EGCG at final concentrations(5,10,20,40,80 μmol/L).The inhibitory rates of the cells were detected by MTT assay and IC50 was calculated.BGC-823 cells were treated for 24 h with EGCG at final concentrations(12,24,48 μmol/L).The cells were collected and total RNA was extracted by Trizol method and VEGF mRNA was detected by quantitative real-time PCR.The medium was also collected and assessed for VEGF levels by ELISA.Results The inhibitory rate of the BGC-823 cells was significant positive correlation between the inhibitory rates and concentrations of EGCG(r=0.977,P0.05).The experimental IC50 of cell proliferation was(47.94±3.26) μmol/L.BGC-823 cells were treated for 24 h with EGCG at final concentrations(12,24,48 μmol/L) and mean values of the expression of VEGF mRNA were 0.63±0.05,0.25±0.02,0.16±0.01.Significant negative correlation was found between the expression of VEGF mRNA and concentrations of EGCG(r=-0.855,P0.05).Mean values of the expression of VEGF in the mediums were(677.32±41.08),(545.16±28.17),(256.06±11.02) ng/L.Significant negative correlation was found between the expression of VEGF and concentrations of EGCG(r=-0.991,P0.05).Conclusion EGCG can inhibit BGC-823 cell proliferation and the antiangiogenic properties of EGCG which was accompanied by decreases in the expression and secretion of VEGF.
