摘 要:用平板透明圈法对高温平菇(Pleurotus ostreatus)、金针菇(Flammulina velutipes)、香菇(Lentinus edodes)等担子菌菌种进行筛选。选取高温平菇作为诱变出发菌株进行复合诱变处理并进行初筛。选取透明圈半径与菌落半径比值较大的菌株接种至发酵培养基进行复筛。最终筛选出一株产β-葡聚糖酶能力较强的高温平菇突变茵株P32,并进行产酶条件的优化。结果表明:最佳碳源比为麸皮:β-葡聚糖:麦秸粉=9:1:1,氮源为4g/L(NH_4)_2CO_3,起始pH值5.0,培养温度32℃。该菌株在优化条件下产β-葡聚糖酶的能力最强,酶活力达到60.08 IU/mL。Pleurotus ostreatus, Flammulina velutipes and Lentinus edodes and so on, were screened by transparent zones method. Pleurotus ostreatus as initial strain was treated by multiple mutagensis and then the mutants with the bigger ratio of transparent zones radius to colony radius were chosen for further screening. At last, the Pleurotus ostreatus P32 producing high ^-glucanase activity was obtained. The results showed that the optimum carbon sources were bran, β-glucan and barley straw powder in the ratio of 9 : 1 : 1 ; the optimum nitrogen source was 4 g/L (NH4)2 CO3; the optimum initial pH and temperature were pH 5.0 and 32℃, respectively. Under the optimized conditions, the β- glucanase ability of Pleurotus ostreatus P32 reached as high as 60.08 IU/mL.
