摘  要:从11种待选脱色介质中筛选出330(OH)型树脂对纳豆激酶发酵液进行脱色研究。结果表明:(1)330(OH)型树脂对纳豆激酶发酵液中的色素为优吸型吸附,其动力学模型符合扩散方程,其动力学拟和方程为-1n(1-F)=0.0223t+0.0511,R^2=0.9978,其中F=Q_t/Q_e,Q_t为脱色时间为t时330(OH)型树脂对色素的吸附量,Q_e为330(OH)型树脂对色素的平衡吸附量。(2)在静态条件下,当pH 7.0,树脂与发酵液用量比为15 g(树脂):40 mL(发酵液)时,脱色200 min,其脱色率为90%,酶活保留率为97.67%,纯化倍数3.09;(3)利用该树脂进行动态脱色实验,当pH 7.0,流速为1.0 BV/h时,脱色5 h,用pH 7.0的磷酸缓冲液淋洗0.9 BV,将脱色液和淋洗液合并,其脱色率为90%,酶活保留率为98%,纯化倍数为3.41。For decolorization of nattokinase fermentation liquor, resin 330(OH) was selected from 11 different decolorizing mediums. The results showed that (1)Resin 330 (OH)could effectively adsorb the coloring matters in nattokinase fermentation liquor. The kinetic model accorded to diffusing equation well. The kinetic Folynomial equation was -ln( 1 - F) = 0. 0223 t + 0.0511, R2= 0. 9978, in which t was decolonizing time. (2)Under static condition, the proportion of resin and fermentation liquor was 15 g: 40 mL at pH 7.0 for 200 min. The decolorization effidency was 90%, the activity of enzyme was 97.67 and the purification multiple was 3.09. (3)The dynamic condition consisted of pH 7.0, flow-velodty of 1.0 BV/h and operation time 5 h. The decolorizing liquor was mixed with eluant of phosphate buffer at pH 7.0 by flow-velocity 0.9 Bv. The total decolorization efficiency was 90 %, the activity of enzyme was 98 % and the purification multiple was 3.41.
