核心家系5-羟色胺转运体基因启动子区多态性与自杀未遂的关联分析

摘　　要：目的:探讨5-羟色胺转运体(Serotonin transporter,5-HTT)基因启动子区多态性(Serotonin transporter gene-linkedpolymorphic region,5-HTTLPR)位点与自杀未遂之间关联系。方法:以重庆地区汉族人群中自杀未遂核心家系作为研究对象,应用聚合酶链反应技术(Polymerase chain reaction,PCR)检测5-HTTLPR位点多态性与自杀未遂之间的分子遗传学联系进行了以家系为基础的基于单体型的单体型相对风险(Haplotype based haplotype relative risk,HHRR)分析及连锁不平衡(Trans-mission disequilibrium test,TDT)分析。结果:86个核心家系258个家系成员的5-HTTLPR基因型和基因频率在患者组与父母组间比较(χ2=0.088,P=0.767;χ2=2.670,P=0.263)无显著性差异。HHRR分析结果显示自杀未遂患者组与虚拟对照组之间的5-HTTLPR(χ2=0.26,P>0.05)基因频率无显著性差异。多等位基因TDT统计分析5-HTTLPR与自杀未遂间未发现存在连锁不平衡(χ2=2.67,P>0.05)。在57个有精神病自杀未遂核心家系中有47个杂合子父母纳入5-HTTLPR位点的分析,其中有25个S等位基因传给了患病子女,有12个L等位基因传递给了患病子女(χ2=4.57,P<0.05)。结论:汉族人群中5-HTTLPR多态性位点与有精神病自杀未遂者间存在连锁不平衡。Objective：To explore the association between attempted suicide and polymorphism of serotonin transporter gene-linked polymorphic region（5-HTTLPR）.Methods：Suicide attempts core families were recruited from the Han population in Chongqing.Using the technique of polymerase chain reaction（PCR）and restriction fragment length polymorphism techniques.A family-based link-age relative risk based on haplotype and disequilibrium analysis were conducted.Results：No significant difference of genotypic frequence of 5-HTT gene polymorphism in the promoter region（5-HTTLPR）between the patients and the parents from 258 family members of 86 core families（χ2=0.088,P=0.767;χ2=2.670,P=0.263）.the HHRR（χ2=0.26,P〉0.05）and the TDT（χ2=2.67,P〉0.05）of multiple allele did not show association between polymorphism of 5-HTTLPR and attempted suicide.There were 47 heterozygous parents of 5-HTTLPR in 57 psychiatric suicide attempts core families,25 S alleles and 12 L alleles of which transmitted to the suffered children（χ2=4.57,P〈0.05）.Conclusion：It is association between polymorphism of 5-HTTLPR and attempted suicide with psychiatric diseases in the Han people.
