摘  要:目的:研究武汉汉族新生儿中肺表面活性物质蛋白D(SP-D)基因多态性及其与支气管肺发育不良(BPD)易感性的相关关系。方法:采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)对218例武汉汉族新生儿SP-D相关位点进行基因型检测,同时进行基因测序验证结果。结果:SP-DM et11Thr三种基因型TT、TC、CC频率分别为10.5%、49.1%、40.4%,等位基因T和C频率分别为35.1%和64.9%。SP-DA la160Thr三种基因型GG、GA、AA频率分别为54.6%、39.4%、6.0%,等位基因G和A频率分别为74.3%和25.7%。与对照组比,SP-DM et11Thr和SP-DA la160Thr基因多态性与BPD发生率无明显关联(P>0.05)。结论:SP-DM et11Thr和SP-DA la160Thr基因型和等位基因频率存在种族差异性;SP-DM et11Thr和SP-DA la160Thr基因多态性与BPD发生率无明显关联。AIM: To analyze the genetic polymorphisms of surfactant protein D(SP-D) and to investigate the association between polymorphisms and bronchopulmonary dysplasia(BPD) in Han racial Wuhan newborns.METHODS: Genotyping of SP-D polymorphisms was performed by polymerase chain reaction and restriction fragment length polymorphism analysis(PCR-RFLP) in 218 Han racial Wuhan newborns.The method of gene sequencing was used to validate the results.RESULTS: The frequencies of TT,TC,CC of SP-D Met11Thr were 10.5%,49.1% and 40.4%,respectively,and the allele frequencies of T and C were 35.1% and 64.9%,respectively.The frequencies of GG,GA,AA of SP-D Ala160Thr were 54.6%,39.4% and 6.0%,respectively,and the allele frequencies of G and A were 74.3% and 25.7%.SP-D Met11Thr and SP-D Ala160Thr displayed no significant association with BPD as compared with control group.CONCLUSION: The differences in SP-D Met11Thr and SP-D Ala160Thr polymorphisms are significant among several ethnic groups.Polymorphisms of SP-D Met11Thr and SP-D Ala160Thr display no significant association with BPD as compared with control group.
