摘 要:以APMP木浆造纸污泥为生物质原料,采用酿酒酵母与重组大肠杆菌KO11组合,同步糖化共发酵生产燃料乙醇,并对其工艺条件进行优化。结果表明:最适反应条件为温度38℃,底物浓度50 g/L,混合菌接种量为6%(体积分数),纤维素酶用量25 FPU/g底物,酿酒酵母与重组大肠杆菌KO11接种比例1?1(细胞干质量初始浓度分别为1.0 g/L和0.3 g/L)。在此工艺条件下发酵72 h,乙醇产率可达0.115 g乙醇/g底物,发酵效率达到理论值的42.31%。扫描电镜分析显示造纸污泥经同步糖化共发酵后,纤维含量减少,纸浆纤维被降解成小块碎片。Primary sludge from APMP pulp paper mill sludge was used as biomass material to produce fuel ethanol with Saccharomyces cerevisiae and E. coli KOll via simultaneous saccharifieation and co-fermentation(SSCF). The SSCF process conditions were then optimized at a temperature of 38℃, with a substrate concentration of 50 g/L, a mixed bacteria inoeulums size of 6 % ( volume fraction), and a cellulase dosage of 25 FPU/g, Saccharomyces cerevisiae and E. coli KOll inoculum ratio of 1 : 1 (the initial concentration of cell dry mass were 1.0 g/L and 0. 3 g/L). With these optimal technologcial conditions, the yield of ethanol reached 0. 115 g/g after a 72 hours incubation, the theoretical fermentation efficiency was 42. 31 % . Scanning electron microscopy (SEM) analysis shows that the paper- making sludge treated by SSCF, the fiber content decreased, pupl fibers were degraded into small fragments.
