摘 要:通过比较了6种大孔吸附树脂D4006、NKA-9、X-5、HPD450、HPD600和AB-8对花椒总黄酮的吸附及脱附性能,筛选出最佳树脂进行动态实验研究,得到优化的纯化条件。结果表明:AB-8型大孔树脂纯化效果最好,其最佳工艺为:上样浓度3.0~4.0mg/mL,上柱pH4.5左右,上柱速度2.5mL/min,洗脱剂最适宜体积分数与洗脱剂用量分别为70%和3BV(柱体积bed volume)。经AB-8型大孔吸附树脂纯化后,花椒黄酮的纯度由51.46%提高到90.25%,总黄酮回收率81.65%。对经AB-8大孔树脂纯化后的花椒黄酮进行高效液相色谱(HPLC)分析发现,黄酮纯度显著提高,其中芦丁含量较高。Adsorption and desorption characteristics of macroporous adsorption resins D4006, NKA-9, X-5, HPD450,HPD600 and AB-8 toward flavones of Zanthoxylum bungeanum were investigated.The best macroporous adsorption resin was chosen to carry out dynamic adsorption experiments, while the purification process was acquired.The results showed that the AB-8 macroporous resin gave the best effect of separation with concent ratio 3.0-4.0mg/mL,the feed solution pH 4.5 and the sampling rate 2.5mL/min, respectively, and the concent ratio of elution was 70% alcohol, with eluant volume 3BV ( bed volume). By this technology the flavonoid content was increased from 51.46% of the crude extract to 90.25% of the purified product and the yield was 81.65%. High performance liquid chromatography (HPLC) analysis result showed that the purity of obtained flavonoids was significantly increased,and the rutin content was higher than other flavonoids.
