摘 要:目的:优化回流萃取、RP-HPLC检测荚果蕨中槲皮素、山奈酚含量的方法,比较不同采收时期荚果蕨营养叶中总黄酮含量,得出荚果蕨营养叶的最佳采收时期。方法:色谱条件:色谱柱Shimadzu C18(250×4.6mm,5μm);流动相为甲醇(A)-0.2%磷酸水(B);采用线性梯度洗脱;流速为1.0mL/min;柱温为30℃;检测波长为360nm;进样量20μL。结果:槲皮素在0.18~4.5μg(r2=0.9992),山奈酚在0.17~4.25μg(r2=0.9994)范围内进样量与峰面积呈良好的线性关系。加样回收率分别为99.6%、99.45%,RSD值为1.90%,0.91%(n=6)。结论:该方法操作简便,结果准确,重现性好,可用于荚果蕨的质量评价。9月份采收的荚果蕨营养叶中总黄酮含量最高,实验结果可用于指导合理采收开发荚果蕨资源。Using the methods of reflux extraction and detection of quercetin and kaempferol in Matteuccia struthiopteris(L.)Todaro by RP-HPLC,the content of fiavonoid in different harvest periods and evaluated the optimal harvest time were compared.The separation of the targets was performed on Shimadzu CL8 (250mm × 4.6mm,5μm)column with the column temperature at 30℃, by gradient elution using methanol-H2Owith 0.2% phosphate acid as the mobile phase at the flow rate of 1.0mL/min,the detective wavelength was set at 360nm.The linear range of quercetin was 0.18-4.5μg ( r2 = 0.9992 ) and kaempferol was 0.17-4.25μg ( r2 = 0.9994 ), with average recoveries of 99.6% ( RSD 1.90% ) ,99.45% ( RSD 0.91% ).The method was simple, accurate and reproducible, can be used for quality control of quercetin and kaempfero in Matteuccia struthiopteris( L.).The contents of flavonoid collected at september was the best. These can guide the reasonable collecting and make the best use of Matteuccia struthiopteris( L. ) resource.
