摘 要:采用响应面法同时优化了玉米黄粉中醇溶蛋白和玉米黄素的提取工艺。在单因素实验基础上,选取乙醇体积分数、提取温度、提取时间为影响因子,以玉米黄素和玉米醇溶蛋白质量为响应值,应用Box-behnken中心组合试验设计建立数学模型,进行响应面分析。得到提取的最优工艺条件为乙醇体积分数83.36%、提取温度62.13℃、提取时间5.79h,此条件下玉米黄素质量为420.1μg,而玉米醇溶蛋白质量为0.6165g。与响应面所得理论值418.9μg和0.6153g十分接近,玉米黄素得率和玉米醇溶蛋白提取率分别达到0.021%、79.03%。该研究为工业化提供了依据。Response surface methodology was applied to optimize the extraction conditions of zeaxanthin and zein synchronously from corn gluten meal.According to single factor investigations,a mathematical model with the extraction content of zeaxanthin and zein as responses to volume fraction of ethanol,duration and temperature was established using Box-Behnken central composite experimental design.This was followed by response surface analysis.The optimal conditions of extraction were obtained by response surface methodology as follows:extracted with 83.36% ethanol for 5.79 h at 62.13 ℃,in which conditions the extraction content of zeaxanthin and zein were as high as 418.9 μg and 0.6153 g.The zeaxanthin yield and the extraction rate of zein were 0.021% and 79.03%.The base of the industrial production of zeaxanthin and zein extracted from corn gluten meal was provided.
