摘  要:研究了以海藻酸钠为载体固定化胰蛋白酶的条件,并考察了固定化酶的最适pH、最适温度、储存时间及反复利用稳定性。结果表明:最适固定化条件为CaC12质量分数为3.0%,海藻酸钠质量分数为3.0%,酶液量与海藻酸钠体积比为1∶2,固定化时间为3.0 h;固定化胰蛋白酶最适pH为9.0,最适温度为60℃,较游离酶分别提高1.0和10℃;4℃下保存30 d,活性保留96%以上;连续反应6次,活性保留52%。The optimum conditions for the immobilized trypsin using sodium alginate as the carrier were studied.The optimal conditions for immobilization trypsin were obtained as CaC12 3.0%(ratio of mass to volume),sodium alginate 3.0%(ratio of mass to volume),trypsin 1.5%(ratio of mass to volume) and 3 h for immobilization reaction.Furthermore,the results showed that the optimum pH value and temperature for immobilization trypsin were 9.0 and 60℃ respectively,which enhanced 1.0 and 10 ℃ compared to that of free enzyme.And the relative activity was still over 96% after 30 days.Moreover,the immobilized trypsin had good operational stability,which could be repeatedly used for 6 times and the relative activity of the trypsin was as high as 52%.
