摘  要:通过补加前体提高酿酒酵母SAM-J5E-4发酵生产S-腺苷-L-蛋氨酸的水平.采用10 L罐发酵,考察在相同的培养条件下,补加不同类型的蛋氨酸前体对S-腺苷-L-蛋氨酸合成的影响.当补加前体D-蛋氨酸和L-蛋氨酸时,S-腺苷-L-蛋氨酸的产量分别为1.53 g/L和4.66 g/L,而未补加前体蛋氨酸的对照仅为0.38 g/L.发酵过程中补加2倍量的DL-蛋氨酸前体对促进S-腺苷-L-蛋氨酸的积累效果最佳,补料方式为流加,补加速率为4 g/(L.h),共流加5 h,在此条件下S-腺苷-L-蛋氨酸的产量为5.28 g/L,生物量达120 g/L以上.以DL-蛋氨酸为前体应用于S-腺苷-L-蛋氨酸的流加发酵过程可明显提高发酵水平.To improve the yield of S-adenosyl-L-methionine(SAM) by feeding methionine as precursor during the fermentation of Saccharomyces cerevisiae SAM-J5E-4,the effects of feeding strategies and the amount of different kinds of methionine on the production of SAM were investigated in 10 L fermentator,respectively.The results indicated that feeding D-or L-methionine during the fermentation,the accumulation of SAM reached 1.53 g/L and 4.66 g/L,in comparison with 0.38 g/L of the control.DL-methionine was the most suitable precursor for the accumulation of SAM.When the precursor amino acid was fed at a rate of 4 g/L/h for 5 h,the maximum yield of 5.28 g/L and more than 120 g/L of biomass were obtained.The feeding of DL-methionine during the fermentation could obviously enhance the productivity of SAM.
