摘  要:目的:探讨桂皮醛对慢性髓细胞白血病细胞株K562生长的影响及其机制。方法:以不同浓度的桂皮醛作用于体外培养的K562细胞,CCK-8法检测细胞增殖活性,流式细胞术检测细胞周期、凋亡率Caspase 3表达。结果:桂皮醛呈时间和剂量依赖性影响K562细胞增殖,使细胞周期受阻于G2/M期,并诱导K562细胞凋亡。K562细胞凋亡过程中,活化的Caspase 3表达明显增加。结论:桂皮醛使K562细胞周期受阻,并使Caspase-3活化诱导细胞凋亡,从而实现对K562细胞的生长抑制。Objective: This study was purposed to investigate effects ofcinnamic aldehyde on the proliferation of chronic myeloid leukemic cell line K562 and its mechanism.Methods:K562 cells were treated by various concentrations of cinnamic aldehyde, The proliferation of K562 cells was detected by CCK-8 assay. Flow cytometry was employed to measure the cell cycle distribution, apoptosis and the expression of active Caspase 3 of K562 cells.Results:Cinnamic aldehyde affected the proliferation and induced the apoptosis of K562 cells in a time- and dose- dependent fashion. Cinnamic aldehyde arrested K562 cells at G2/M. The levels of active Caspase 3 arose in cinnamic aldehyde treated K562 cells. Conclusions:Cinnamie aldehyde inhibited proliferation of K562 cells in vitro via arresting cell cycle and inducing apoptosis. Actived Caspase 3 played an important role in apoptosis ofK562 cells.
