摘 要:目的:研究设计一种新的用于分离与鉴别念珠菌的显色培养基及方法。方法:针对念珠菌特有的酶,设计出配方,应用质控菌株,通过特异性、灵敏度及生理特征方面比较实验,评价其效果。结果:本实验研制的念珠菌显色培养基具有较好的选择性和特异性,菌落特征出现时间早于科玛嘉念珠菌显色培养基,另外,本方法通过底物5-溴-4-氯-3-吲哚基-N-乙酰基-β-D-氨基半乳糖苷和5-溴-4-氯-3-吲哚基-β-D-吡喃葡糖苷反应,可以将都柏林念珠菌与白色念珠菌区分开,同时使白色念珠菌得到进一步确证。结论:新的显色培养基及方法能更有效地用于白色念珠菌及4种相关重要念珠菌的分离与鉴别,并且缩短了检测时间,降低了检测成本,具有较好的应用价值。Objective:To develop a new chromogenic medium for isolation and identification of Candida spp.Methods: The new chromogenic medium was designed base on the characteristic enzyme of Candida spp.By using the reference strains,the sensitivity,specificity and efficiency of the new chromogenic media were compared with that of CHROMagar Candida.Results: The results showed that the selectivity and specificity of the developed chromogenic media were as the same as that of CHROMagar candid,but the colony characteristics appeared earlier in that of CHROMagar,moreover,by adding substrate 5-Br-4-Cl-3-indolyl-N-acetyl-β-D-galactosamine dry and 5-Br-4-Cl-3-indolyl-β-D-pyranoid rkatsiteli grapes reaction,C.albicans can not only be differentiated with C.dubliniensis but also be further confirmed.Conclusion: The new developed Chromogenic medium is effective for isolation and identification of C.albicans and 4 kinds of relative important Candida with short period and low cost and has good application value.
