摘  要:目的探讨雌激素受体GPR30、ERα和PR在甲状腺癌组织中的表达及其相关性。方法选取重庆医科大学附属第一医院病理科2006-2009年101例甲状腺癌组织标本,包括乳头状腺癌83例、滤泡状腺癌9例、髓样癌6例、未分化腺癌3例,患者年龄45.9(9~93)岁;采用免疫组化S-P法检测甲状腺癌组织中GPR30、ERα和PR的表达,分析其与肿瘤临床病理特征之间,以及相互间的差异。结果甲状腺癌组织中GPR30、ERα和PR主要表达于细胞质,少量表达于细胞核,阳性表达率分别为78.2%、85.1%、79.2%;除在≥45岁年龄组患者肿瘤中ERα表达率高于〈45年龄组外(r=-0.2383,P〈0.05),GPR30和ERα的表达与患者年龄、性别、肿瘤大小、淋巴结转移、病理类型、TNM分期等临床病理因子及患者MAC IS预后评分无统计学差异(P〉0.05);GPR30与ERα(r=0.2518)、PR(r=0.3207)的表达呈正相关。结论雌激素受体GPR30和ERα共同参与甲状腺癌的发生、发展,可能是潜在的治疗靶点。Objective To evaluate the expression of G-protein coupled receptor 30(GPR30),estrogen receptor α(ERα) and progesterone receptor(PR) in thyroid carcinoma(TC) tissues,and to study their association with thyroid carcinoma.Methods Immunohistochemistry(S-P) assay was used to detect the expression of GPR30,ERα and PR in the paraffin-embedded TC tissue specimens from 101 patients(age at 45.9,9-93 years).The specimens were collected in our hospital from 2006 to 2009 and included 83 cases of papillary adenocarcinoma,9 cases of follicular adenocarcinoma,6 cases of medullary carcinoma and 3 cases of undifferentiated carcinoma.The association of GPR30,ERα and PR expression with TC clinicopathologic features and the correlation among them were analyzed.Results GPR30,ERα and PR were expressed mainly in cytoplasm and rarely in cell nuclei,with positive expression rates of 78.2%,85.1% and 79.2%,respectively.Their expression displayed no significant association with clinicopathological determinants(age,sex,tumor size,lymph node metastasis,pathologic subtype and TNM stage) and MACIS prognostic score(P0.05),except that ERα had a higher positive expression rate in patients aged 45 years and older than in patients aged less than 45 years old(r=-0.2383,P0.05).GPR30 expression was positively correlated with ERα expression(r=0.2518) and PR expression(r=0.3207).Conclusion As estrogen receptors,GPR30 and ERα both contribute to the development of TC and,therefore,may be potential therapy targets.
