摘  要:糖化酶是一种被广泛应用的重要的工业用酶,本实验从酒曲中筛选出一株糖化酶高产菌株,该菌菌丝初为白色,后变为土黄色,菌落呈厚绒状,质地紧密。土黄色球形分生孢子着生在顶囊的小梗上。经菌落、细胞形态观察以及基于18SrDNA序列的系统发育关系分析,鉴定该菌株为曲霉属黑曲霉变种,与黑曲霉的序列相似性为99.77%,命名为黑曲霉Aspergillus niger M1。同时与实验室现有黑曲霉菌株Aspergillus nigerAS3.4309进行糖化酶活力比较,为其酶活力的3.38倍,有进一步应用开发价值。Glucoamylase is an important industrial biocatalyzer and broadly used in bioindustry. In this paper, a glucoamylase production strain was isolated from mouldybran samples. The myeelial color is white at first, and then becomes yellowish brown, velvet - like colonies are thick and close texture. The yellow sphere conidia is on the small stalk of the acrocyst. The identification of colony morphology and cell morphology and the phylogenetic analysis based on 18SrRNA sequence shows that the strain is a variant of Aspergillus niger and the sequence similarity is 99.77%. The strain was identified and denominated Aspergillus niger M1. At the same time, the glucoamylase activity of Aspergillus niger M1 was investigated and compared with Aspergillus niger AS3. 4309 preserved in our laboratory, the enzyme activity of the former was 3.38 times than that of the latter. Therefore, Aspergillus niger M1 is a portential strain for industry development in the future.
