搜风祛痰中药对AngⅡ诱导人脐静脉内皮细胞NF-KBmRNA及iNOSmRNA表达的影响

摘　　要：[目的]观察搜风祛痰法对血管紧张素Ⅱ致体外培养人脐静脉内皮细胞(HUVECs)NF-KB mRNA及iN-OSmRNA表达的影响,阐明搜风祛痰法的中药调控NF-KBmRNA与iNOSmRNA在AS过程中可能性的作用机理。[方法]制备兔活心汤含药血清。采用酶消化法培养HUVECs2-3代用于实验,采用RT-PCR法测定内皮细胞NF-KBmR-NA及iNOS mRNA。随机分为5组:正常对照组,AngⅡ组,低浓度中药血清+AngⅡ组中浓度中药血清+AngⅡ组高浓度中药血清+AngⅡ组。[结果]活心汤可抑制AngⅡ导致的内皮细胞损伤,减少NF-KBmRNA及iNOSmRNA的表达。[结论]活心汤可抑制AngⅡ所致的HUVECs损伤,保护血管内皮功能。[Objective]To observe the way of appling dispel the wind and searching phlegm to angiotensin II to in vitro human umbilical vein endothelial cells（HUVECs） NF-iNOSmRNA expression mRNA KB and influence.Clarify display of traditional Chinese medicine to the way of appling dispel the wind and searching phlegm of KBmRNA and iNOSmRNA-NF process in AS the possibility of mechanism.[Methods]The serum containing metabolic ingredients of Huoxin Decoction was taken from rabbits.HUVECs2-3 were cultured in vitro by collagenase digestive method and NF-KB mRNA and iNOSmRNA were identified with RT-PCR method.HUVECs were randomly divided into five groups：normal control group,Ang II group,low concentration drug serum and Ang II group,middle concentration drug serum and Ang II group and high concentration drug serum and Ang II group.[Results]Huoxin Decoction could inhibit the injury of endothelial cells and decrease the expression of NF-KB mRNA and iNOSmRNA.[Conclusion]Huoxin Decoction could inhibit the injury of HUVECs induced by Angiotensin Ⅱ to protect vascular endothelial.
