酶催化制备富含多不饱和脂肪酸的磷脂酰丝氨酸及其抗肿瘤活性研究

摘　　要：以亚临界1,1,1,2-四氟乙烷(R134a)作为非水介质,以磷脂酶D催化鱿鱼卵磷脂与L-丝氨酸的酰基转移反应制备富含多不饱和脂肪酸(EPA/DHA)磷脂酰丝氨酸(PS),探讨了亚临界R134a体系条件对酶促反应的影响。采用四氮唑蓝还原法(MTT法),研究所制备的EPA/DHA磷脂酰丝氨酸对人宫颈癌细胞(Hela细胞)和结肠癌细胞(Caco-2细胞)的体外抗肿瘤活性。结果表明,磷脂酶D在亚临界R134a中显示出高活性,在6 MPa、40℃、加水2.4 mL、反应240 min的条件下,酰基转换反应制备EPA/DHA磷脂酰丝氨酸的转化率为84.77%;制备的EPA/DHA磷脂酰丝氨酸对Hela细胞和Caco-2细胞增殖有显著抑制。200μg/mL PS作用于Hela细胞72 h,可使其存活率下降30%;200μg/m LPS作用于Caco-2细胞72 h,可使其存活率下降40%。Phospholipase D-catalyzed transphosphatidylation from phosphatidylcholine of egg of squid Symplectoteuthis oualaniensis to produce n-3 polyunsaturated fatty acid-riched Phosphatidylserine was performed in subcritical 1,1,1,2-tetrafluroethane（R134a）.The impacts of medium parameters such as pressure,temperature and water addition on the enzymatic preparation were discussed;Then anti-tumor in-vitro activities of prepared EPA/DH-riched Phosphatidylserine by MTT assay were investigated.The results indicated that Phospholipase D exhibited high activity in subcritical R134a and conversion was 84.77 % under these conditions： 6 MPa,40℃,water addition 2.4 mL,reaction time 240 min.The prepared PS showed distinct suppressant activity to cell Hela and Caco-2.The survival rates of cell Hela and Caco-2 declined by 30% and 40% respectively,after treatment with 200 μg/mL prepared PS for 72 h.
