摘  要:目的检测PDGF-BB及其受体PDGFR-β在NSCLC中的表达,探讨其表达与NSCLC淋巴管生成及淋巴道转移的关系。方法采用免疫组化SP法检测PDGF-BB及其受体PDGFR-β在113例NSCLC癌组织中的表达水平,分析它们的表达与临床病理参数之间的关系;应用D2-40进行淋巴管染色,计数NSCLC中微淋巴管密度(LMVD);分析PDGF-BB及其受体与微淋巴管密度之间的关系,探讨其在NSCLC淋巴管生成及淋巴道转移中的作用。结果 PDGF-BB主要表达在NSCLC癌细胞的胞膜和胞浆中,其阳性表达率为63.7%(72/113);PDGFR-β主要表达在NSCLC癌巢周围的间质细胞的细胞质中,阳性表达率为61.0%(69/113);PDGF-BB在NSCLC癌细胞中的表达与间质中PDGFR-β的表达明显相关。PDGF-BB、PDGFR-β表达水平与患者的年龄、肿瘤大小、组织类型、分化程度均无关,而与TNM分期、淋巴结转移有关。PDGF-BB阳性表达组其LMVD显著高于阴性表达组(16.5817±5.4556对11.4689±5.5679,P<0.05);PDGFR-β阳性表达组其LMVD显著高于阴性表达组(15.7937±5.6383对11.6917±6.3862,P<0.05)。结论 PDGF-BB及其受体PDGFR-β在NSCLC上的阳性表达可以促进肿瘤淋巴管的生成及淋巴转移,针对这些信号通路的抑制剂的研发将为抑制肿瘤淋巴转移带来新的治疗前景。Objective To examine the expression of platelet-derived growth factor-BB(PDGF-BB) and its receptor PDGFR-β and to investigate their relationship to lymphangiogenesis and lymphatic metastasis in non-small-cell lung cancer(NSCLC).Methods Immunohistochemical method was used to determine the lymphatic microvessel density(LMVD) and the expression of PDGF-BB and its receptor PDGFR-β in 113 specimens from patients with NSCLC and 20 normal lung tissue samples.Results PDGF-BB was highly expressed mainly in the membrane and cytoplasm of lung tumor cells.The expression rate was 63.7%(72/113).PDGFR-β was highly expressed mainly in the cytoplasm of the cells of tumor stroma.The expression of PDGF-BB and PDGFR-β was significantly higher in NSCLC tissues than in normal lung tissues(both P0.05,respectively).The expression of PDGF-BB and PDGFR-β in NSCLC tissues had no correlation to age,gender,tumor size,histological type and differentiation grade.However,the expression of PDGF-BB and PDGFR-β was positively correlated to TNM stage and lymph nodes metastasis.LMVD was significantly higher in groups with positive expression of PDGF-BB and PDGFR-β than in groups with negative expression,and high PDGF-BB or PDGFR-β expression was significantly more common with lymph node metastasis.Conclusion The high expression of PDGF-BB and its receptor PDGFR-β can promote lymphangiogenesis in tumor and metastasis to lymph nodes,which indicates that PDGF-BB and PDGFR-β may play an important role in lymphangiogenesis and lymphatic metastasis in NSCLC.
