摘 要:1引言α-茄碱是变绿发芽后的马铃薯中含量最高的一种生物碱,具有抗肿瘤、抑菌、强心、镇痛等[1]多种功效。目前对马铃薯中α-茄碱的高效液相色谱检测法(HPLC)报道较少,并存在重现性较差、目标峰难分开等问题。A method was established for the determination of α-solanine in potato by high performance liquid chromatography.The α-solanine was extracted from potato by refluxing extraction.The effects of different mobile phase composition,concentration,concentration ratio,flow speed,column temperature and other factors on α-solanine separation were studied.When the mobile phase was a mixture of acetonitrile and 0.4% phosphoric acid aqueous solution,flow speed was 0.8 mL/min,temperature was 30 ℃,detection wavelength was set at 210 nm,α-solanine could be separated well.The calibration of α-solanine was in good linearity in the range of 0.16-0.80 g/L.The average recovery was 96.3% with relative standard deviation of 1.3%(n=6).
