摘 要:利用吸附、交联的原理,以壳聚糖、聚乙烯醇(PVA)、戊二醛等为材料,通过冻干技术制备了冻干-PVA-壳聚糖固定化酯酶。并通过单因素优化实验,确定了固定化酶生产的适宜条件:壳聚糖浓度3%、PVA浓度3g/L、酶液固定化载体体积质量比5∶2(mL∶g),戊二醛浓度0.4%,交联时间30min,此时固定化酶活力回收率为73.8%。固定化酶在60℃,pH值为7.0的反应体系中活性最大,使用5次后,固定化酶载体形状基本没变化,活性保留了71%,表明冻干-PVA-壳聚糖固定化酶具有良好的操作稳定性。According to adsorption and crosslinking principles, chitosan, PVA, glutaraldehyde etc were used as material to prepare the freeze-PVA-chi- tosan immobilized esterase through the lyophilization technique. The optimal conditions of immobilized enzyme production were obtained by single factor experiment as followed: chitosan 3%, PVA 3g/L, the ratio of enzyme liquid volume to immobilized carrier weight 5:2 (ml: g), glutaraldehyde 0.4% and crosslinking time 30min, Under the above conditions, the recovery rate of immobilized enzyme was 73.8%. The optimal reaction conditions were as followed: temperature 60℃ and pH value 7.0. The basic shape of immobilized enzyme did not change and its activity retained 71% after five times utilization. These experiments suggested that the lyophilization-PVA-chitosan immobilized enzyme had good operating stability.
