摘 要:研究了一种快速检测大肠菌群的细胞培养瓶剂法,即以国标法中的乳糖胆盐发酵培养基为基本营养成分,经浓缩、干燥后制成快速检测瓶剂,待检样品滴入细胞培养瓶内,培养24 h,依据培养瓶盖上产气指示试纸颜色变化及瓶内溶液颜色变化,确定可疑阳性样品,并对其进行氧化酶实验,验证大肠菌群的存在。实验对细胞培养瓶法进行了大肠菌群的重复性及实际样品检测,并与国标法进行比较。结果显示:细胞培养瓶法与国标法所测结果符合率达96%,样品检测报告时间只需24 h,比国标法减少48 h,2种方法所测结果在统计学上无显著性差异(P>0.05)。细胞培养瓶法简单、快速、准确,利于基层监测机构和生产企业对食品中大肠菌群的快速检测。A cell culture bottle method was developed for rapid detection of coliforms in food samples. In this study, lactose bile salt medium were concentrated and dried for bottle testing agent. According to the color change of the gas test strip and the solution in cell culture bottle, suspected positive samples could be determined, and then be verified the presence of coliforms in suspected samples through oxidize and microscopic examination. The results of repetitive tests and actual samples detection were compared with national standard method. A high agreement (96%) was found between the cell culture bottle method and national standard method on 50 samples. The overall assay time of cell culture bottle method only needed 24 h, which reduced 48 h than that of national standard method. The statistical analysis showed that there werenl significant difference (P 〉0.05 ) between the examination results for these methods. As a simple, rapid and accurate detection method, cell culture bottle could meet the needs of the primary monitoring agency and manufacturers for rapid detection of coliforms in food.
