摘 要:[目的]从富含油脂的土壤中寻找高产脂肪酶的真菌,以期为扩大脂肪酶的菌源提供材料。[方法]从上海市、河北省唐山、江苏省兴化、安徽省合肥地区土样中,经富集培养、平板筛选得到22株脂肪酶产生菌株,通过复筛得到1株脂肪酶活力较高真菌。根据《常见与常用真菌》对菌株进行鉴定,并研究不同条件对菌株产酶的影响。[结果]从富含油脂土壤中分离得到1株产脂肪酶能力较高菌株,根据菌落群体及个体形态,查阅相关鉴定手册,初步确定该菌株为黑曲霉。菌株产酶条件研究表明:其最适产酶条件为温度30℃,初始pH 7,装液量20%,最适碳源与氮源分别为淀粉和蛋白胨。[结论]为脂肪酶产生菌以及脂肪酶的工业化生产与应用奠定了基础。[Objective]The study aimed to seek for the fungi with high yield of lipase from the greasy soil,so as to provide the material for expanding the fungi source of lipase.[Method]22 strains of fungi producing lipase were isolated by enrichment culture and flat screening methods in soil samples collected from Shanghai City,Tangshan City of Hebei Province,Xinghua area of Jiangsu Province,Hefei area of Anhui Province.A stain with higher activity of producing lipase was isolated by re-screening methods.The strain was identified according to "The Common and Common-used Fungi" and the effects of different conditions on the strain producing lipase were studied.[Result]A stain with higher ability of producing lipase was isolated from the greasy soil.The strain was identified as Aspergillus niger according to groups characteristic and individuals forms of colonies and the relevant appraisal manual.The study on the conditions of producing enzyme for the strain showed that the optimal conditions of producing enzyme were as follows: the temperature for fermentation of 30 ℃,the initial pH of 7,the medium volume of 20%,and the optimum carbon and nitrogen sources were starches and peptones,resp.[Conclusion]The study laid a foundation for lipase producing bacteria and industrial production and application of lipase to some extent.
