摘  要:目的建立HPV-16阳性的宫颈上皮永生化细胞(H8细胞株)和宫颈癌细胞(CaSKi细胞株)胞膜蛋白双向电泳图谱,寻找特征性的差异蛋白点。方法提取两种细胞胞膜蛋白质进行双向凝胶电泳(2-dimensronal gel electrophore-sis,2-DE),建立电泳图谱,然后筛选出差异点进行基质辅助激光解吸飞行时间串联质谱(matrix-assisted laser desorption/i-onization time-of-flight tandem mass spectrometry,MALDI-TOF-MS/MS)分析,鉴定出蛋白质。结果获得了分辨率和重复性均较好胞膜蛋白2-DE图谱,并分析找出了13个的差异蛋白点,对其中3个显著差异点进行质谱分析和初步鉴定。结论建立了H8和Caski细胞胞膜蛋白2DE图谱,并初步发现LRC8D、EDAR、MTX1蛋白在宫颈癌前病变和宫颈癌细胞胞膜中存在差异表达,为宫颈癌及癌前病变的诊断及癌变机制提供了帮助。Objective To identify differentially expressed membranous protein spots of from human immortalized endocervical cell(H8) and cervical cancer cell(Caski) associated with cervical carcinogenesis by two-dimensional electrophoresis(2-DE) analysis.Methods The membranous proteins of H8 cell and Caski cell were separated by means of two-dimensional gel electrophoresis,then the selected differential protein spots were analyzed by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF-MS) and SWISS-PROT database.Results 13 differentially expressed proteins spots from these membranous proteins in H8 cells and Caski cells were further identified by MALDI-TOF-MS analysis and 3 of them were successfully identified and characterized.LRC8D(Leucine-rich repeat-containing protein 8D)、EDAR(Tumor necrosis factor receptor superfamily member EDAR precursor)、Metaxin-1 were significantly up-regulated in membranous proteins in H8 cells.Conclusion There differential expressions of membranous proteins of may be valuable for studying mechanisms of cervical carcinogenesis,or as diagnostic markers and therapeutic targets for cervical cancer.
