摘  要:目的:探讨hTERT反义寡核苷酸(ASODN)抑制CM-319细胞端粒酶活性对脊索瘤细胞的细胞周期及增殖的影响。方法:用脂质体介导hTERT正义寡核苷酸(SODN)和反义寡核苷酸(ASODN)转染CM-319脊索瘤细胞株72小时后,MTT法检测细胞活性,流式细胞仪检测细胞周期及细胞凋亡率变化。结果:hTERT的ASODN转染细胞的端粒酶活性及细胞生长都明显受到抑制。流式细胞仪检测显示:ASODN组细胞出现早期凋亡峰,细胞阻滞于S期,而hTERT的SODN则无此作用。ASODN组细胞凋亡率为16.01%,而hTERT的SODN组为2.3%。结论:端粒酶hTERT为靶点的反义寡核苷酸可明显抑制脊索瘤细胞的增殖,且具有促凋亡作用。Objective:To investigate the effect of inhibiting telomerase activity by hTERT antisense oligodenxynucleotide on proliferation of chordoma cell line CM319.Methods: Human chordoma cell line CM319 was transfected with hTERT antisense oligodenxynucleotide(ASODN) and sense oligodenxynucleotide(SODN),mediated by lipofectamine.MTT was used to examine cell activity.The cell cycle and the rate of apoptosis were determined by flow cytometry 72 hours after treatment.Results: The telomerase activity and the proliferation were markedly depressed in CM319 cell which was transfected with hTERT ASODN.The cell cycle was arrested in S phase,and early apoptotic peak was showed by flow cytometry.The rate of apoptosis was 16.01% while the control group was 2.3%.Conlusion: ASODN targeting hTERT can inhibit the proliferation of chordoma cell line CM319 and induce apoptosis.
