脑益康对高胆固醇血症致Aβ增高大鼠的作用研究

摘　　要：目的观察脑益康对高胆固醇血症致Aβ增高大鼠的作用并分析其作用机制。方法高胆固醇饲料喂饲大鼠建立高胆固醇血症模型,脑益康灌胃,Y电迷宫测试,测定血脂、血流变及脑海马区Aβ表达。结果与模型对照组相比,脑益康高剂量组大鼠电刺激尝试次数显著减少(P<0.01),血清总胆固醇显著降低(P<0.01),大鼠脑海马区Aβ表达积分光密度显著降低(P<0.01)。结论脑益康可改善大鼠学习、记忆,其机制与降低血脂水平、改善血液流变学、减少脑Aβ生成有关。Objective To investigate the effects of Naoyikang on β-amyloid content in rat model with hypercholesterolemia and its mechanism. Methods The rat model of hypercholesterolemia was established by feeding high cholesterol diet,Naoyikang was given by gavage to rats,the behavior changes of rats were tested by Y-maze,the content of β-amyloid was measured by immunohistochemistry and western blotting methods,the content of serum cholesterol and low density lipoprotein-cholesterol were measured by automatic biochemical analyzer,and maximum aggregation index and blood viscosity were also observed. Results Compared with model group,the number of attempts in Y-maze test was significantly reduced in high dose group of Naoyikang（P0.01）,the serum cholesterol was significantly decreased（P0.01）,and the low density lipoprotein-cholesterol and maximum aggregation index were decreased（P0.05）.With immunohistochemistry and Western blot,the integral optical density of Aβ in hippocampus of rats,the average optical density were decreased significantly（P0.01）. Conclusion Naoyikang can significantly improve learning and memory ability,and the mechanism is in correlation with reducing the content of Aβ in hippocampus,decreasing serum cholesterol and low density lipoprotein-cholesterol level,promoting blood circulation as well as reducing brain β-amyloid biogenesis.
