白血病患者20例染色体断裂点研究

摘　　要：本研究观察了20例白血病患者外周血淋巴细胞染色体断裂情况，结果表明：①白血病患者染色体断裂点发生率高于正常人；②断裂点增多主要是由于脆性位点表达所致；③脆性位点表达的部位与肿瘤相关断点、癌基因之间有较大一致性；④脆性住点表达的种类与白血病类型之间无明显特异关系。The chromosornal breakpoints in Iymphocytes were sdudied in 20 patients withIeukemia. Specimens of peripheral blood from 13 cases of ANLL, 7 cases of ALL' werecultrued in medium 199 plus 5% FCS and compaired wlth 20 normal controls: The aver-'age chromosomal breakpoint rate of the patients (13. 2F4%) is significantly higher thanthat of the controls. Out of 62 kinds of G-banded chromosomal breakpoints, 40 were lo-cated at the same band with fragile sites ; 28 with cancer breakpoints and 13 with onco-gene locations. An overall consistency of fragile sites with cancer breakpoints as well asoncogene locations difl e:tist,but we did not find any close relation between fragile sitesand the group of subgroup of leukemia. Most of the chromosomal breakpoints ofleukemia are fragile sites. The increased expression of fragile sites may reflect the insta-bility of chromosomes which might be one of the causes of leukernia.
