摘  要:目的探讨普洱茶水提物对巨噬细胞分泌肿瘤坏死因子-α(Tumor necrosis factor-α,TNF-α)的影响。方法以佛波酯处理THP-1细胞48 h,分化得到巨噬细胞,以其为炎症细胞模型,用不同浓度的普洱茶水提物(125、250μg/ml)与终浓度为100 EU/ml的脂多糖的混合物作用于巨噬细胞,ELISA法检测细胞培养液中TNF-α的含量。结果普洱茶水提物能有效抑制巨噬细胞炎症因子TNF-α的分泌,且浓度为250μg/ml的普洱茶水提物对TNF-α分泌的抑制作用强于浓度为125μg/ml的普洱茶水提物,呈剂量依赖性。结论普洱茶水提物对巨噬细胞炎症因子TNF-α的分泌具有抑制作用。这种效果与已知的绿茶抗炎的主要成分EGCG无关。Objective To investigate the effect of Pu-erh tea extract on secretion of tumor necrosis factor-or (TNF-α) by maerophages. Methods Macrophages were differentiated from THP-1 cells after treatment with phorbol ester and used as the model of inflammatory ceils, which were treated by the mixture of lipopolysaccharide (LPS) at a final concentration of 100 EU/ml with Pu- erh tea extract at various concentrations (125 and 250 μg/ml) and determined for TNF-α content in cell culture by ELISA. Restilts Pu-erh tea extract effectively inhibited the secretion of TNF-α by macrophages. However, the inhibitory effect of 250 μg / ml Pu-erh tea extract was stronger than that of 125 Ixg/ml extract. Conclusion Pu-erh tea extract showed dose-dependent inhibitory ef- fect on secretion of TNF-α by maerophages, and the effect was unassociated with EGCG, one of the major anti-inflammatory components of green tea.
